| 目的:探讨葫芦素脂质体滴鼻剂对ANIT诱导的黄疸型HB大鼠模型的干预作用及其可能机制。方法:采用熔融法制备葫芦素脂质体滴鼻剂,并建立ANIT诱导的黄疸型HB大鼠模型。将大鼠随机分为正常对照组、模型对照组、熊去氧胆酸组、提取物灌胃组(800 mg/kg、400 mg/kg)与提取物滴鼻组(100 mg/kg、50 mg/kg),连续给药7天。检测各组大鼠的血清肝功能指标、免疫组化分析PXR和LXR的表达,探讨葫芦素脂质体滴鼻剂的干预作用及可能机制。结果:与模型对照组相比,提取物灌胃组显著改善TBIL、TBA、GGT、DBIL等肝功能指标。其中提取物滴鼻组对ALB显示出特有上调效应。免疫组化结果显示,正常对照组PXR和LXR表达极低,模型对照组表达明显升高,各给药组PXR和LXR表达均有所改善,提取物滴鼻组改善较为显著。结论:葫芦素脂质体滴鼻剂可通过上调ALB,调节PXR和LXR的表达,改善ANIT诱导的黄疸型肝损伤,为黄疸型HB的精准化治疗提供新的策略与分子靶点。 |

| Objective: To investigate the interventional effect and potential mechanism of Cucurbitacin liposomal nasal drops on an ANIT-induced rat model of icteric hepatitis B. Methods: Cucurbitacin liposomal nasal drops were prepared using the melting method, and an ANIT-induced rat model of icteric hepatitis B was established. Rats were randomly divided into a normal control group, a model control group, a ursodeoxycholic acid group, an extract gavage group (800 mg/kg, 400 mg/kg), and an extract nasal drop group (100 mg/kg, 50 mg/kg). Administration continued for 7 consecutive days. Serum liver function indicators were measured, and immunohistochemical analysis was performed to assess the expression of PXR and LXR, exploring the interventional effect and potential mechanism of Cucurbitacin liposomal nasal drops. Results: Compared with the model control group, the extract gavage group significantly improved liver function indicators such as TBIL, TBA, GGT, and DBIL. The extract nasal drop group specifically showed an upregulating effect on ALB. Immunohistochemical results indicated that PXR and LXR expression was very low in the normal control group, significantly increased in the model control group, and improved in all treatment groups, with the extract nasal drop group showing more notable improvement. Conclusion: Cucurbitacin liposomal nasal drops can ameliorate ANIT-induced icteric liver injury by upregulating ALB and modulating the expression of PXR and LXR, providing new strategies and molecular targets for the precise treatment of icteric hepatitis B. |
