      目的:探讨鱼腥草乙醇提取物(HCT)对鼻咽上皮细胞炎症反应的抑制作用及其对核因子κB(NF-κB)信号通路的影响。方法:采用人鼻咽上皮细胞(NP69),以脂多糖(LPS)诱导炎症模型。细胞分别经不同浓度HCT(0.05、0.1、0.2 mg/mL)预处理1 h后给予LPS刺激。通过MTS法检测细胞活力;酶联免疫吸附试验(ELISA)检测细胞上清液中肿瘤坏死因子-α(TNF-α)、白细胞介素-6(IL-6)、白细胞介素-8(IL-8)水平;逆转录聚合酶链反应(RT-PCR)分析TNF-α、IL-6、IL-8 mRNA表达;蛋白质免疫印迹(Western blot)法检测IκBα磷酸化及NF-κB p65核转位;荧光素酶报告基因法测定NF-κB启动子活性。结果:与模型组比较,HCT能显著提高LPS诱导降低的细胞活力(P<0.05);显著抑制LPS诱导的TNF-α、IL-6及IL-8分泌及其mRNA表达(P<0.05或0.01);显著抑制IκBα磷酸化及NF-κB p65核转位(P<0.05或0.01);显著下调NF-κB启动子活性(P<0.05或0.01)。结论:HCT可能通过抑制NF-κB信号通路活化,阻断IκBα降解及NF-κB核转位,从而抑制鼻咽上皮细胞促炎细胞因子产生,为中药治疗咽部炎症性疾病提供实验依据。

      Objective: To investigate the inhibitory effect of Houttuynia cordata ethanol extract (HCT) on the inflammatory response of nasopharyngeal epithelial cells and its impact on the nuclear factor kappa B (NF - κ B) signaling pathway. Method: Human nasopharyngeal epithelial cells (NP69) were used to induce an inflammatory model with lipopolysaccharide (LPS). Cells were pretreated with different concentrations of HCT (0.05, 0.1, 0.2 mg/mL) for 1 hour and then stimulated with LPS. Detect cell viability using MTS method; Enzyme linked immunosorbent assay (ELISA) was used to detect the levels of tumor necrosis factor - α (TNF - α), interleukin-6 (IL-6), and interleukin-8 (IL-8) in the cell supernatant; Reverse transcription polymerase chain reaction (RT-PCR) was used to analyze the mRNA expression of TNF - α, IL-6, and IL-8; Western blot was used to detect I κ B α phosphorylation and NF - κ B p65 nuclear translocation; Determination of NF - κ B promoter activity using luciferase reporter gene assay. Result: Compared with the model group, HCT significantly increased the cell viability induced by LPS (P<0.05); Significantly inhibited the secretion and mRNA expression of TNF - α, IL-6, and IL-8 induced by LPS (P<0.05 or 0.01); Significant inhibition of I κ B α phosphorylation and NF - κ B p65 nuclear translocation (P<0.05 or 0.01); Significantly downregulated NF - κ B promoter activity (P<0.05 or 0.01). Conclusion: HCT may inhibit the activation of the NF - κ B signaling pathway, block the degradation of I κ B α and NF - κ B nuclear translocation, thereby suppressing the production of pro-inflammatory cytokines in nasopharyngeal epithelial cells, providing experimental evidence for the treatment of pharyngeal inflammatory diseases with traditional Chinese medicine.
